Sulphation of a cell surface glycoprotein from Volvox carteri. Evidence for a membrane-bound sulfokinase working with PAPS.

چکیده
Sulphated glycoproteins have been implicated in the control of cell growth and differentiation: fibronectins [ 1,2]; glycoproteins in developing sea urchin embryos [3]; and a surface glycoprotein from V&ox curteri [4]. This V&ox glycoprotein, of 185 000 app. M, [4] appears to be involved in the control of the differentiation of the organism during embryogenesis. Although PAPS or APS must be involved as sulphate donor in the synthesis of such molecules, the incorporation of sulphate from PAPS into a sulphated glycoprotein has been demonstrated only in [5]. It was therefore of interest to partially characterize the sulphate-transfer system operative in Volvos.

Sulphation-desulphation of a membrane component proposed to be involved in control of differentiation in Volvox carteri.
In the developing asexual embryo of Y&XX carteri the differentiation into somatic and reproductive cells is seen at the division from 32-64 cells. At this stage, 16 cells of the 32celled embryo undergo unequal cleavage, forming 16 large reproductive cells [ 1,2]. In sexual embryos developing under the influence of an inducer the differentiating cleavage is delayed in the male strain up to the f...
متن کاملReporter genes and highly regulated promoters as tools for transformation experiments in Volvox carteri.
The multicellular alga Volvox is an attractive model for the study of developmental processes. With the recent report of successful transformation, regulated promoters as well as reporter genes working in this organism are now required. The Volvox genes encoding arylsulfatase and the extracellular glycoprotein ISG are strictly regulated. The former is transcribed only under conditions of sulfur...
متن کاملPurification and partial characterization of a glycoprotein sexual inducer from Volvox carteri.
A method is described for the isolation and purification of the sexual inducer produced by male colonies of the colonial green alga Volvox carteri. The procedure yields a highly purified preparation of the inducer. The biologically active principal is demonstrated to be a glycoprotein with a molecular weight of 32,000.
متن کاملA volvox inversionless mutant highlights the importance of the extracellular matrix in morphogenesis.
Volvox is a multicellular spherical green alga that is closely related to the more widely studied unicellular Chlamydomonas. Volvox is a fascinating creature that serves as a useful model system for investigating the evolution of multicellularity and the regulation of cell differentiation (reviewed in Kirk and Nishii, 2001). An adult Volvox, called a spheroid, consists of several thousand small...
متن کاملA phosphodiester bridge between two arabinose residues as a structural element of an extracellular glycoprotein of Volvox carteri.
The sulphated glycoprotein SSG 185 is the monomeric precursor of a highly aggregated structural element in the extracellular matrix of the multicellular green alga Volvox carteri. A phosphodiester of arabinose was isolated from a saccharide fragment of SSG 185. The structure of this phosphodiester was investigated by methylation analysis, 13C-NMR, photometric methods and enzymatic assays and id...
